Documentation | The myelin sheath is a multi-layered membrane, unique to the nervous system,
that functions as an insulator to greatly increase the velocity of axonal
impulse conduction [1]. The P0 glycoprotein, absent in the central nervous
system [1], is a major component of the myelin sheath in peripheral nerves:
it comprises a large extracellular N-terminal domain, a single trans-
membrane (TM) region, and a smaller positively-charged intracellular
domain. It is postulated that P0 is a structural element in the formation
and stabilisation of peripheral nerve myelin [2], holding its characteristic
coil structure together by the interaction of its positively-charged domain
with acidic lipids in the cytoplasmic face of the opposed bilayer, and by
interaction between hydrophobic globular `heads' of adjacent extracellular
domains [1].
MYELINP0 is a 6-element fingerprint that provides a signature for the P0
family of myelin proteins. The fingerprint was derived from an initial
alignment of 5 sequences: motifs 1-4 were drawn from conserved regions
spanning most of the extracellular domain; motif 5 encodes the TM region;
and motif 6 includes the region encoded by PROSITE pattern MYELIN_P0
(PS00565), which lies in the intracellular domain. Two iterations on OWL23.2
were required to reach convergence, at which point a true set comprising 6
sequences was identified. A single partial match was also found, HSMYELIN,
which matches all but the last motif as a result of an insertion mutation
in the gene.
An update on SPTR37_9f identified a true set of 6 sequences, and 4
partial matches.
